1 Tirzepade derivaves Tirzepade (CAS-No. 2023788-19-2) is a glucose-dependent insulinotropic polypepde and glucagon-like pepde-1 receptor agonist. According to the custom in the field, the sequence of amino acids is writen from N-terminus (le) to C-terminus (right), any substuents to an amino acid side chain are indicated in brackets () to the right of the amino acid concerned, and the amino acids are in L-configuraon unless otherwise specified. The “H-“ at the N-terminus indicates a free amino group, the “- NH2” at the C-terminus indicates a carboxamide group and the “-OH” at the C-terminus indicates a carboxylic acid group. Posions along the pepde chain may be given by numbers starng with 1 at the N-terminus and may be indicated in superscript to the right of the amino acid concerned. Posions within the side chain substuent of an amino acid start with 1 for the moiety atached to the side chain of the amino acid and may be indicated with a dot in superscript to the right of the amino acid concerned. For example, in case of a Lys at posion 20 as the amino acid, the moiety atached to the epsilon-nitrogen atom of Lys is numbered with “moiety20.1”. For praccal reasons, a “moiety” covers an amino acid as well as a non- amino acid, which is bound via an amide bond, e.g., a faty acid. For the sake of brevity, some or almost all or all posion indicators may be le out, without changing the informaon. Formula I is Tirzepade’s molecular structure expressed in the three-leter-code with an individual numbering of all amino acids: H-Tyr1-Aib2-Glu3-Gly4-Thr5-Phe6-Thr7-Ser8-Asp9-Tyr10-Ser11-Ile12-Aib13-Leu14-Asp15-Lys16-Ile17- Ala18-Gln19-Lys20(AEEAc20.1-AEEAc20.2-gamma-Glu20.3-19-carboxy-nonadecanoyl20.4)-Ala21- Phe22-Val23-Gln24-Trp25-Leu26-Ile27-Ala28-Gly29-Gly30-Pro31-Ser32-Ser33-Gly34-Ala35-Pro36-Pro37- Pro38-Ser39-NH₂ (I) Formula II is Tirzepade’s molecular structure expressed in the three-leter-code without an individual numbering of all amino acids: 2 H-Tyr-Aib-Glu-Gly-Thr-Phe-Thr-Ser-Asp-Tyr-Ser-Ile-Aib-Leu-Asp-Lys-Ile-Ala-Gln-Lys(AEEAc- AEEAc-gamma-Glu-19-carboxy-nonadecanoyl)-Ala-Phe-Val-Gln-Trp-Leu-Ile-Ala-Gly-Gly-Pro- Ser-Ser-Gly-Ala-Pro-Pro-Pro-Ser-NH₂ (II) In both formulae I and II, AEEAc represents the amino acid 8-amino-3,6-dioxa-octanoic acid. Formula I and formula II above describe the same compound, i.e., Tirzepade. In the following, Tirzepade is compound No. 100. It will be understood that a pepde such as Tirzepade or a Tirzepade derivave may be present as a salt with various counter-ions both at side chain funconal groups and terminal funconal groups. A numbering of an amino acid is used in the following in case a reference is made to a posion of the amino acid. Tirzepade derivaves are of interest as pharmaceucal compounds, as some may be expected to have similar physiological effects as Tirzepade. As such, they might be suitable for treatment of metabolic diseases, for example to improve glycemic control in adults with type 2 diabetes or for weight management. A synthesized Tirzepade derivave allows an evaluaon of its physiological effects including its applicaon in respecve in-vitro test system. Moreover, small amounts of Tirzepade derivaves may be comprised in a pharmaceucal composion containing Tirzepade as a desired drug substance. For example, the material obtained from a manufacturing process of Tirzepade, which is used as the drug substance in a preparaon of a pharmaceucal composion containing Tirzepade as a desired drug substance, contains already small amounts of Tirzepade derivaves. A synthesized Tirzepade derivave supports an analycal determinaon of the Tirzepade derivave assumed to be in a pharmaceucal composion containing Tirzepade as a desired drug substance or in the material obtained from a manufacturing process of Tirzepade. These are reasons, why synthesis of Tirzepade derivaves is of interest. Overview of Tirzepade derivaves Some of the Tirzepade derivaves can be assigned to certain categories due to their way of differing from Tirzepade. A way of differing can be a missing amino acid, i.e., a deleon (des). Similarly, a way of differing can be an addional amino acid, which is a doubling of what should be solely one amino acid, i.e., an addion (endo). Another way of differing can be an exchange 3 of one amino acid with a different amino acid, i.e., a substuon. Another way of differing can be a change of an amide group into a carboxylic acid group, i.e., a deamidaon. Another way of differing can be a missing of one or more amino acids at the N-terminal end of Tirzepade, i.e., a truncaon. There are cases, where one and the same difference of a Tirzepade derivave from Tirzepade can be assigned to two different categories. For example, a deamidaon of Gln to Glu is also a substuon of Gln by Glu. For example, a truncaon is a specific deleon, i.e., a deleon at the N-terminal end. Compounds, which are Tirzepade derivaves, are listed in th...
